Parijoto fruit (Medinilla speciosa Blume) is an epiphytic shrub with a height of 0.45-1.2 meters. Parijoto fruit contains secondary metabolites of flavonoids, anthosionins, glycosides and others which have pharmacological effects as antioxidants, antidiabetic, anticancer, antibacterial, anti-inflammatory. Parijoto plant growth is influenced by temperature, humidity, rainfall and altitude where it grows. This study aims to determine the total flavonoid content in Parijoto fruit from Kudus and Semarang districts. This type of research is an experimental laboratory research using UV-Vis spectrophotometry method using quercetin and rutin as a comparison. The total flavonoids of Parijoto fruit extract from Kudus Regency and Semarang, respectively, with a quarcetin ratio of 81.60 mgQE/g and 76.48 mgQE/g, while with a routine comparison of 79.33 mgRE/g and 73, respectively. 06 mgRE/g. The total flavonoids of the two extracts showed quite strong results, but were significantly different as evidenced by the value of the paired sample t-test between the two extracts with a significance value (2-tailed) <0.05.
